Two-stage synthesis of CTP using 20 mM cytidine as substrate. The reaction was initiated at 50 °C in 50 mM PB buffer (pH 5.0) containing 2 mM ATP, 5 mM polyP6, 20 mM MgCl2, and pure enzyme to generate UMP. After 2.0 h of the first-stage reaction, the pH and temperature of the solution were adjusted, and the remaining enzyme and substrate were added to produce CTP. All measurements were carried out in triplicates
