PsPIWI-RE possesses RNA-guided DNA cleavage and DNA-guided RNA cleavage activity. A DNA cleavage assays of PsPIWI-RE were examined in the presence of 21 nt guides with different 5′-terminal modifications, which were as follows: 5′ phosphorylated (5′P) DNA, 5′hydroxyl (5’OH) DNA, 5′ phosphorylated (5′P) RNA, and 5′ hydroxyl (5′OH) RNA. A 78 nt single stranded DNA (ssDNA) with a 3′ FAM label was the target sequence. No Guide: reaction without guides. “–”: reaction without guides and PsPIWI-RE. B RNA cleavage assays of PsPIWI RE were examined in the presence of 21 nt guides with different 5′-terminal modifications, which were as follows: 5′ phosphorylated (5′P) DNA, 5′ hydroxyl (5′OH) DNA, 5′ phosphorylated (5′P) RNA, and 5′ hydroxyl (5′OH) RNA.
